Why do multiphoton imaging with 1040 nm fixed wavelength lasers?
1040 nm is very well suited for multiphoton microscopy for several reasons:
-
It falls into the NIR transparency window of biological tissues spanning between 650-1300 nm
-
Scattering at 1040 nm lower than at e.g. 800 nm, which allows deeper imaging (Rayleigh scattering (λ-4) dependency).
-
Abundancy of fluorophores excitable at ~1040 nm.
-
SHG signal at 520nm is easy to detect.
